For high(est) mastery points and high(est) achievement points you need to play all expansions and all seasons and (nearly) all content in the game. That is quite a long term goal by itself. I would say prioritize the expansions (and their follow-up seasons) chronologically. If you want convenience, get the core-tyria-autoloot mastery first.

If there are none left in a map you know it's not a real one you're looking for.
